Sažetak
Membranska se tehnologija uspješno primjenjuje za pročišćavanje bromelaina, proteaze dobivene iz ananasa. Međutim, fluks permeata i separacija enzima u postojećim sustavima nisu optimalni, uglavnom zbog svojstava smjese, poput viskoznosti. Stoga smo u ovom radu prethodnom obradom enzimima i dijafiltracijom u dvostupanjskom membranskom procesu poboljšali učinkovitost pročišćavanja i koncentriranja enzima bromelaina, dobivenog iz ekstrakta mješavine krutog otpada od ananasa. Provedena je prethodna obrada ekstrakta otpada pektinazom, celulazom ili njihovom mješavinom i uspoređen je njihov učinak na smanjenje prividne viskoznosti. Dijafiltracija je uvedena u drugom stupnju ultrafiltracije, kad su praćeni sljedeći parametri: fluks permeata, te iskorištenje i čistoća enzima. Prethodnom obradom ekstrakta pektinazom postignuto je smanjenje prividne viskoznosti od 12 %, čime je fluks povećan za 37-38 % u oba stupnja ultrafiltracije, a postignuto je i bolje iskorištenje enzima u prvom stupnju ultrafiltracije. Uvođenjem dijafiltracije u drugom stupnju procesa zadržane su visoke vrijednosti fluksa, i enzim je pročišćen 4,4 puta (u prethodnom radu je pročišćen 2,5 puta), no za to je potrošena veća količina otapala. Rezultati ovog istraživanja pokazuju da je moguće poboljšati fluks permeata i separaciju bromelaina smanjenjem viskoznosti smjese prethodnom obradom enzimima i dijafiltracijom. Obje se metode mogu primijeniti u industrijskim membranskim postrojenjima za proizvodnju bromelaina.
