The distribution of restriction fragment length polymorphism (RFLP) across the Zagorje turkey and hybrid chicken ovalbumin locus has been mapped in DNA genome isolated from blood of 30 animals, representing four phenotype varieties. Using the same primers, polymerase chain reaction (PCR) was used for amplification of the ovalbumin gene of Zagorje turkey and chicken genomic DNA. In addition to the size of amplified ovalbumin gene, the order of restriction fragments was determined by comparing a map of the EcoRI, HaeIII, PvuII and AvaI sites in Zagorje turkey and chicken. The lengths of PCR products derived from 40 turkey DNA genomes were 2.47 kb without significant differences. Two polymorphic restriction sites in PCR products such as AvaI site at position 2150 bp and PvuII at 1718 bp allows genotyping of Zagorje turkey and chicken. Our results suggest that the part of the ovalbumin gene is identical in all four Zagorje turkey phenotypes at the location of Hrvatsko Zagorje. The comparison between chicken and Zagorje turkey ovalbumin genes showed high polymorphic diversity.
