Pharmacological properties of 1,3-acetone bis -(4-hydroxyiminoformyl pyridinium) dibromide (S-101)

Abstract
In experiments on the anaesthetized dog it was found that intravenous injection of 30 mg/kg S-101 (1,3-acetone bis /4-hydroxyiminoformyl pyridinium/ dibromide) produces a blood pressure decrease of about 25% of the initial value, but does not affect the respiratory function. This dose of S-101 has no effect on the hypotensive action of injected acetylcholine, hut successfully prevents the blood pressure drop produced by electric stimulation of afferent vagus nerve. The blockade of vagus impulses due to S-101 was also observed after the application of adrenaline whose pressor effect was slightly intensified by the oxime. The repeated application of S-101 and TMB-4 in short time intervals produced the total apnea and a critical decrease in blood pressure. However, the total dose of S-101 necessary to block the respiratory movement was 50% larger than the total dose of TMB-4. Respiratory blockade which is probably the primary cause of death could not be prevented by nikethamide. A relatively short artificial respiration applied on time produced the return of spontaneous respiration and normalization of blood pressure.
