Biochemical response of lettuce to treatments with plasma activated water and organic fertilizers/biostimulants

In the context of increasing demands for sustainable agriculture and high-quality food production, this study investigates the effects of plasma-activated water (PAW) and organic fertilizers supplemented with biostimulants on the synthesis of secondary metabolites in lettuce (Lactuca sativa L.) cv. Maradona. The objective of this study was to evaluate the impact of PAW treatments, as well as the application of organic fertilizers in combination with biostimulants, on the accumulation of secondary metabolites in lettuce. The experiment was conducted using a two-factorial design at the experimental site of the University of Slavonski Brod, under both greenhouse and open-field conditions. PAW was applied at two dosages (P1 and P2), while organic fertilizers (S, P, B) were combined with biostimulants (C, O, S) in multiple treatment combinations. The data were analyzed using correlation analysis and analysis of variance (ANOVA). The results indicated that phenolic compounds are the primary contributors to antioxidant activity. In treatments involving organic fertilizers and biostimulants, antioxidant activity was positively correlated with proline content (r = 0.72), whereas no significant correlation with total phenolics was observed. In contrast, PAW treatments exhibited a very strong correlation (r ≥ 0.98) between phenolic content, flavonoids, and antioxidant activity, particularly under greenhouse conditions. Lettuce grown under open-field conditions exhibited higher concentrations of phenolics and flavonoids compared to greenhouse-grown plants. Under field conditions, PAW (P1) significantly increased phenolic acids, flavonoid content, and antioxidant capacity, whereas in greenhouse conditions, the higher dose (P2) exerted an inhibitory effect. Among the tested treatments, the CS and SC combinations induced the highest biochemical responses, achieving the greatest accumulation of phenolics (2.5580 mg GAE/g fresh weight) and flavonoids (14.0920 mg CE/g fresh weight in CS). The PO treatment resulted in the highest proline accumulation (2.447 µmol proline/g fresh weight). Overall, the findings suggest that plasma-activated water and selected biostimulants in combination with organic fertilizers can be strategically employed to enhance the nutritional quality of lettuce. However, their effectiveness is strongly influenced by environmental stress conditions.
