Determination of lead in blood and urine by visual identification of the lower sensitivity limit on a chromatogram

Abstract
A simple method for the determination of lead in blood and urine by thin layer chromatography (TLC) without use of a special equipment is described. A mineralized residue of 0.5 ml blood or 5 ml urine is dissolved in 6N hydrochloric acid and mounted in progressively increasing quantities to a thin layer of microcrystal cellulose. Lead is separated from other metals by development of chromatograms in a solvent system: methanol - isopropanol - water - 6M HCl - cone. HNO3 - hydrogen peroxide (3 - 2 - 4 - 0.5 - - 0.5 - 1). After the spots are detected with 4-(2-pyridylazo) resorcinol (PAR) lead is determined by means of the lower sensitivity limited as the last hardly visible spot on the chromatogram.
