DESTRUCTIVE METHODS FOR VIABILITY TEST OF AMBROSIA ARTEMISIIFOLIA L.

Sažetak

In order to estimate the germination dynamic of common ragweed (Ambrosia artemisiifolia L. ) it is necessery to determinate percentage of viable seed in the soil seed bank. Since is impossible to visually distinguish dormant from germinating and dead seed, it is necessary to conduct some additional tests. Destructive methods to test seed viability are: germination test + tetrazolim (TTC), individual tetrazolium (TTC) and crush test. Each of these methods have some limitations and since tetrazolium and crush test are in certain part subjective methods, the aim of this research was to: I) compare the viability results of destructive methods and II) to determine if TTC test and crush test can give reliable and repeatable results. Laboratory research was conducted on common ragweed seed which was collected in September 2014 in Popovača. In experiment I seed was tested through germination test + TTC test, individual TTC test and crush test. Experiment II was seperated on reliability of testing TTC and crush test. In TTC test in experiment a difference in visual perception of 2 evaluators and the same evaluator in time interval between 2 evaluations (60') was tested, and in experiment b aim was to determine possible differences between evaluations of the same evaluators for both halves of same seed, and to compare evaluations from 1st evaluator who evaluated one half of the seed with another half which was evaluated by 2nd evaluator. In crush test, difference between two evaulators was determined where one seed half seed was evaluated by 1st evaluator and other half by 2nd evaluator. Results showed that there were no significant difference in seed viability between germination test + TTC test, individual TTC test and crush test. First evaluation in TTC test from the same evaluator in time lag from 60 minutes compared to another one, was not significantly changed. Also, the results of the comparison of the first and second evaluations of both evaluators indicate that they evaluated the viability of the seed in a similar way, confirming the reliability and repeatability of the TTC test (experiment a). The halves of the same seed of common ragweed were not evaluated the same by both evaluators (experiment b). Crush test gave objective results in seed viability estimation beacause with this test it was not determined significant difference in assesment of viability between both evaluators.
