Purification of Hyodeoxycholic Acid and Chenodeoxycholic Acid from Pig Bile Saponification Solution Using Column Chromatography

Sažetak

The purpose of this study was to purify hyodeoxycholic acid and chenodeoxycholic acid from pig bile saponification solution using column chromatography. The optimal resin was selected through static adsorption experiments. Loading, washing, and elution conditions were optimized. Hyodeoxycholic acid and chenodeoxycholic acid were further purified through crystallization. The best resin was found to be CG161M macroporous resin. After optimization, the sample was loaded at a volume flow of 3 BV h–1 to
adsorb 5 BV, washed with 40 % ethanol, eluted with 45 % ethanol and 60 % ethanol, respectively. This resulted in purities for hyodeoxycholic acid of 70.34 % and 66.21 % for chenodeoxycholic acid with yields of 86.48 % and 90.57 %, respectively. Crystals of hyodeoxycholic acid with a purity of 91.04 % and chenodeoxycholic acid precipitate with a purity of 80.28 % were obtained through crystallization. Hyodeoxycholic acid and chenodeoxycholic acid can be purified using CG161M resin, and subsequent crystallization yields high-purity hyodeoxycholic acid.
