Study of automated epifluorescent microscopy for evaluation of raw milk bacteriological quality

Abstract
The applicability of BactoScan 8000, instrument for automated total bacteria count determination was tested as method of evaluation raw milk if supplier's price was dependent on hygienic quality. The BactoScan estimates were compared to colonies number determined using standard plate count method (IDF Standard 100 B: 1991). After logarithms transformation, between BactoScan (y) and CFU method (x) the regression equation y = 0.916 x +0.714 and the residual standard deviation sy/x = 0.26 log units were calculated. The correlation coefficient between two methods was 0.95 (n = 701). The repeatability of the Instrument (precission, sr) at different impulse levels varied from 0.02 tog units to 0.11 log units when colonies' number was <100,000 CFU/ml. The established limit of detection for P = 0.95 was ± 0.517 log BSC impulses/ml.
