Hydrolysis of thyroglobulin and extraction of ionized aminoacids from the thyroid tissue

Abstract
The influence of the hydrolysis time on the amount of unhydrolyzed thyroglobulins wax examined. Hydrolysis was carried out with trypsin, pancreatin and papain. The influence of the tune of extraction with n-butanol on the amount of the extracted iodized amino acids was also tested. The results obtained by chromatographic separation of pure hydrolyzate and butanol extract of the hydrolyzate were compared. Experiments were performed on the thyroid glands of white rats. The rats were injected 50 µc of radioactive iodine (J-131). Chromatographic separations were carried out in solutions n-butanol : ethanol : 2N NH2 (5 : 1 : 2) and n-butanol : acetic aoid : water (12 : 3 : 5). For the detection of marked compounds on the chromatograph the autoradiographic method was used. Quantitative measurements were performed with a scintillation counter.
