Abstract

Immunohistochemical staining is a method for detection of presence and localization of specific antigens in cells and/or tissues. It is based on the basic principle of immunoreactions: binding of specific antibody to a certain antigen. Development of immunohistochemical staining methods started during the 1930s and, in a number of variations, they are still used today both in research and routine diagnostic procedures.
This paper offers presentation of immunohistochemical methods development and gives an example of protocol optimization for detection of BACH2 antigen in human secondary lymphoid tissue. Most important steps of the protocol are discussed (section preparation, antigen demasking and specific antibody incubation conditions). Optimal protocol for immunohistochemical staining for BACH2 antigen is given and can be used in any routine hematopathological laboratory.
