Hydrolysis of Some Monomethylcarbamates in Human Sera

Abstract
The rate of hydr.olysis of 1-naphthyl-N-methylcarbamate (sevin),
3-isopropylphenyl-N-methylcarbamate (IPFC) and 2-isopropoxyphenyl-
N-methylcarbamate was measured 1in sera of 12 individuals.
SevJn was hydrolysed 3.1 times faster than IPFC, while 2-isopropoxyphenyl-
N-methylcarbamate did not hydrolyse under given experimental
conditions. The Michaelis constants (K m) and the maximal
rates of hydrolysis (V) of seviin and IPFC were deter.mined. Both,
K m and V varied from one serum to the other. For each serum
Vsevin was 2.9 times higher than VrPFc. The Michaeli s constants
for both compounds varied w.ithin the sa me range.
SeVlin and IPFC were not hydrolysed by the serum fraction
IV-1. Serum fractiOIIl V hydrolyse d sevin approximately 4 times
faster than !PFC. Native s erum and serum fraction V did not alter
their activity upon incubation at 53° Cup to 3 and 6 hr., respectively.
It is suggested that the same enzyme(s) hydrolyse sevin and !PFC ·in
human sera.
