Abstract

Pepti des are acti ve regulators and informati on carriers in biological systems,
which makes them interesti ng for pharmaceuti cal purposes. Pharmaceuti cal pepti des can
be manufactured in many ways but pepti de producti on via solid phase pepti de synthesis is
commonly used because of fi nal product safety.
Liquid chromatography and mass spectrometry have been successfully employed in pharmaceuti
cal industry both as separated or hyphenated techniques for quanti tati ve analysis,
characterizati on of impuriti es, structure analysis of pepti des, and for pepti de/protein modifi
cati on determinati on. Mass spectrometry ionizati on techniques MALDI (Matrix-assisted laser
desorpti on/ionizati on) and ESI (Electrospray ionizati on) are primary ionizati on techniques
for converti ng nonvolati le biomolecules like pepti des or proteins, into a gas phase.
ESI-MS is oft en coupled on-line with liquid chromatography or capillary electrophoresis
while MALDI-TOF MS is used as off -line technique. Also, MALDI is suitable for analysis of
water insoluble pepti des. Additi onally, it is possible to use MALDI in the off -line mode for
analyzes of protein samples aft er protein digesti on and purifi cati on which have been separated
either by gel electrophoresis or capillary zone electrophoresis.
