Sažetak
The blood levels of HI-6 after intramuscular administration were found to be dose related. Doses of 10, 20, 30 and 50 mg per kg of HI-6 produced plasma concentrations of 20.2, 39.1, 48.5 and 78.6 µg per ml, respectively. PAM-Cl in doses of 20 and 50 mg per kg produced respective plasma concentrations of 17.1 and 42.7 µg per ml. With HI-6 the time necessary to reach peak concentrations varied from 5 min. for a 10 mg dose to 25 min. for a 50 mg dose. The half-times of oxime elimination from blood were 26.8 min. for HI-6 and 18.4 for PAM-Cl. As an increase in the dose did riot increase the half-time, it is assumed that the first-order kinetics of elimination was involved. The volume of distribution for PAM-Cl was 4-5 times larger than for HI-6, indicating that HI-6 diffuses through membranes or into tissues more slowly than PAM-Cl when Hl-6 was given in doses of 10 mg per kg intramuscularly at one-hour intervals, it was possible to maintain blood levels of more than 4 µg per ml for several hours. The biological half-life, calculated from blood plasma values after oral administration of 200 mg per kg was 101.9 min. for HI-6, and 180.5 min. for PAM-Cl. The maximum concentration of HI-6 in plasma after oral administration was about 11 µg per ml. Oral administration of the oxime may produce smaller therapeutic effect due to lower absorption of HI-6.
