Comparison of accuracy and reliability of two methods for the determination of lead in blood

Sažetak
Two sensitive, fast and easily performed analytical procedures for the determination of lead in whole blood have been compared: spectrophotometric dithizone method and voltametric method. Both methods were developed and published separately by some of the present authors. The spectrophotometric »monocolour« dithizone method for the determination of small amounts of lead in blood (5 ml) was published in 1952 and in 1971 it was improved and adapted for small blood samples (2 ml). In both methods the dithizone reaction is performed at a rather high pH value (10.5). By electrochemical deposition followed by anodic oxidation of lead on the dropping mercury electrode and subsequent voltametric determination using a d. c. polarographic technique a sensitivity of 5 µg Pb/100 ml in a sample of only 0.2 ml can be achieved. Ten blood samples of humans with no known previous occupational exposure to lead were analyzed. The results obtained with the voltametric method were considerably higher than those obtained with the dithizone method in the concentration range up to 10 µg Pb/100 ml. At higher concentrations the two methods showed a satisfactory agreement.
