Medium as Isolation Factor for Intact Etioplasts

Abstract

This paper describes parallel investigations carried out on various media of saccharose and sorbitol used in pea etioplast isolation.
On the basis of results obtained it can be concluded that entirely etioplasts are difficult to obtain in the pea plant.
In the largest number of etioplasts isolated in a saccharose medium there occur smaller or larger vacuoles, frequently together with other changes. Etioplasts isolated in a sorbitol medium will considerably swell, particularly the thylakoids which appear in the form of vesicles.
No vacuolization was noticed after adding ficoll to the saccharose medium, but there comes forth a large number of etioplasts with various extensions, which disappear after a time. Etioplasts isolated in a sorbitol-ficoll medium show similar changes alongside the swelling of thylakoids.
Considerable improvement in the preservation of etioplasts was achieved by adding 1—5 M Na2EDTA.
One of the possible reasons facilitating the described changes on isolated etioplasts is the presence of the peripheral reticulum, which makes the etioplasts sensitive to external conditions.
