RECENT DATA ON THE MECHANISM OF FUMONIZIN B1 NEUROTOXICITY

Sažetak

Fumonisins are a group of mycotoxins produced by Fusarium spp. moulds, predominately by F. verticilloides (formerly F. moniliforme) mould. From their discovery in 1980s till now, more than 15 types of fumonisins have been isolated and from all isolated fumonisins, FB1 is the most prevalent one. Fumonisins are found all around the world on various commodities, but they mostly contaminate maize and maize based food and feed. In domestic animals consumption of fumonisinscontaminated feed causes equine leukoencephalomalacia (ELEM) in horses, and pulmonary oedema in pigs. To experimental animals fumonisins are found to be nephrotoxic, hepatotoxic and carcinogenic. In certain regions of the world where maize is staple food (as Central America and South Africa) development of several
human diseases, as such primary liver cancer, oesophageal cancer, and neural tube defect is connected with exposure to fumonisins. However, the link between the consumption of fumonisin-contaminated food and development of mentioned diseases is not confirmed in epidemiological studies. The International Agency for Research on Cancer (IARC) classified the most prevalent fumonisin, FB1 as a possible
carcinogen to humans (group 2B). On cellular level several mechanisms of FB1 toxicity are studied and the most important one is disturbance of sphingolipid metabolism. Since FB1 treatment induced changes in parameters of oxidative stress, oxidative stress was suggested as mechanism of FB1 toxicity. Our recent studies on
cells of neural origin indicate that FB1 may cause cell death by disturbing calcium homeostasis and thus disturbance of calcium homeostasis could be one of possible mechanisms of fumonisins neurotoxicity as well as toxicity.
