Comparison of efficiency of different methods of extracting nematodes from the soil

Abstract
The goal of this research was to determine which method of extracting nematodes is the most efficient and which one enables the cleanest sample. The goal was also to determine which method is best for obtaining the highest number of nematodes after 24 and 48 hours.
Samples were collected in the field of 1700 m2, where potato was previously cultivated. The sampling was performed on March 21, 2011 in the area of Našice. A total of six samples was collected and analyzed. The extraction of nematodes from the soil was performed on Faculty of Agriculture in Osijek, Department for Plant Protection, Chair for Entomology and Nematology by three methods: Seinhorst’s bottle method (Seinhorst, 1956), Cobb’s sieving method (Cobb, 1918) and Baermann’s funnel method (Baermann, 1917).
Baermann’s sieving method obtained the cleanest samples. After 24 hours the count of nematodes was 463 individuals. After 48 hours the count of nematodes did not change significantly. There were 472 nematodes. In Seinhorst’s bottle method, that count amounts 251 individuals after 24 hours. The count of nematodes significantly increased after 48 hours and it was 644 nematodes. Dirty samples were obtained by Cobb’s sieving method. The count of nematodes in the last experiment was 194 individuals after 24 hours, and the count increased to 520 individuals after 48 hours. The total number of extracted nematodes by the sieving method was 714, by the bottle method it was 895 and by the funnel method it was 935 individuals.
It can be concluded that the cleanest samples were obtained by Baermann’s funnel method. Cobb’s sieving method has shown itself to be quite inefficient and complicated. It has been proved that time of extraction affects the number of nematodes, and soil humidity and kind of soil affect the extraction. Baermann’s funnel method has shown itself to be the simplest, fastest (24 hours) and a very effective method.
